You can find the Sanctum Sanctorum in Spider-Man 2 by going to Greenwich. The structure is on the western side of the map and southeast of the Greenwich area. It’s on the border of the Financial District. It’s fairly difficult to spot, but the highlight you want to look out for is its strange purple, orange, and yellow-stained glass.

Unfortunately, Spider-Man doesn’t reference the building at all while staring at it. It would have been neat to get a nugget of information on Doctor Strange in this game’s universe. There is a mission involving it and some of the residents inside the building, however.

Perhaps Insomniac Games could make an intriguing Doctor Strange game after Marvel’s Wolverine.

Are there any other Marvel references in this game?

There are quite a few secrets to be found in Marvel’s Spider-Man 2. For example, you can look at the Stan Lee tribute statue on the Upper West Side and Aunt May’s grave in Harlem. The ultra-tall Avengers Tower also stands proudly on the Upper East Side.

Do you think we’ll ever see a game based on The Avengers from PlayStation? It could be better than Crystal Dynamics’ mostly multiplayer take on the property.

There’s even a reference to the Fantastic Four in the game. There are so many possibilities with this universe Insomniac and PlayStation is making.
